Liquid Culture to Agar Filled Petri Dish

Goal

The goal is to transfer liquid culture onto an agar filled petri dish in order to provide the mycelium with a sterile and nutrient rich enviroment that will aid and speed up growth as well as allow the mycologist to choose a segment with the strongest mycelium to ensure success. 

Materials Required

  • Petri Dishes fill with Agar 
  • Liquid Culture or Spore syringe 
  • Rubbing Alcohol 
  • Still Air box 
  • Gloves 
  • Lighter 
  •  Paper Towels 

Method

1. Gather equpment 

2. Heat oven 150 degrees Celsius (Farenheight) 

4 Wash and dry petri dishes 

5. Wrap 10 petri dishes in foil 

6. Place wrapped petri dishes in oven

Conclusion

My method of pouring and inoculating was subpar because I haven’t done agar work in a while

I had more contaminations than normal and that could be because of

  • Knocking over the petri dishes when adding the parafilm
  • Leaving the petri dishes (without parafilm) in the still air box for 3 days
  • Not shaking the liquid culture syringe properly
